Approved Uses

Acne vulgaris.

Off-Label Uses

Off-label (evidence varies): photoaging/photodamage, post-inflammatory hyperpigmentation, melasma (as part of combination regimens), keratosis pilaris, and flat warts.

Dosage & Administration

Dosing by Condition

Acne vulgaris: apply a thin film to the entire affected area once daily in the evening/at bedtime after gentle cleansing and drying; maximum is once daily.

Initial Dose

Apply a thin film to the affected area once daily in the evening after washing.

Maintenance Dose

Once daily application at bedtime; continue for at least 12 weeks to assess efficacy

Maximum Dose

Apply a thin film once daily (do not apply more than once per day)

Children's Dosage

Not approved for children under 12 years. Ages 12 and above: same as adult dosing - apply thin layer once daily at bedtime

Dose Adjustment Notes

No systemic dose adjustment is needed; if excessive irritation occurs, use a moisturizer and/or reduce application frequency (e.g., every other night) or temporarily stop until tolerated, then restart.

How to Take

Cleanse gently and pat skin completely dry; apply a thin film to the entire acne-affected area (not spot treatment) once daily in the evening/at bedtime; avoid eyes, lips, and mucous membranes; wash hands after application.

Side Effects

Common Side Effects

Application-site irritation including dryness, erythema (redness), scaling/peeling, burning/stinging, and pruritus.

Side Effect Frequency

Very common (>10%): dryness, erythema, scaling, burning/stinging/irritation. Common (1-10%): pruritus, sunburn, skin discomfort. Uncommon (<1%): contact dermatitis, eyelid edema, conjunctivitis, rash/eczema, skin discoloration (hyper/hypopigmentation).

Patient Counseling

Use once daily at night on clean, completely dry skin; apply a thin layer (e.g., pea-sized amount for the whole face) to the entire affected area; avoid eyes/lips/mucosa and broken or sunburned skin; expect dryness/redness/peeling and possible early flare-use a non-comedogenic moisturizer and reduce to every other night if needed; use sunscreen and limit sun exposure; avoid harsh/abrasive or other irritating topical products unless advised; results usually require 8-12 weeks.

No routine monitoring required. Monitor for excessive skin irritation or allergic contact dermatitis

Onset of Action

Improvement is typically seen after 8-12 weeks of consistent use; transient early irritation and/or apparent worsening can occur in the first 2-4 weeks.

Duration of Effect

Benefit is maintained only with continued use; acne commonly recurs after discontinuation.

Half-Life

Unknown/not clinically meaningful with topical use due to negligible systemic absorption.

Bioavailability

Systemic absorption after topical application is minimal; plasma levels are generally very low/undetectable (often below assay quantification limits).

Excretion

For any absorbed fraction, elimination is mainly via biliary excretion with fecal elimination (minor urinary excretion).
